Organization Overview
We study nuclear pore complexes (NPCs), which provide the only known sites for nucleocytoplasmic exchange. Using a variety of techniques and yeast as a model system, we are determining the composition and fine structure of the NPC and relating this to its sites of interactions and reactions with soluble nuclear transport factors. We are also examining associations between NPCs and other components of the nuclear periphery. We hope to gain a better understanding of the part the NPC plays in nuclear regulation and maintenance.
Overview
LINE-1 (L1) retrotransposons are genetic parasites that proliferate in our genomes through a 'copy-and-paste' mechanism. L1-encoded proteins (ORF1p, ORF2p) preferentially assemble with the L1 RNA that encodes them, producing ribonucleoprotein (RNP) intermediates that are imbued with pathogen associated molecular patterns. These assemblies are poorly understood and heterogeneous in composition, incorporating many other host-derived RNAs and proteins that form abundant cytoplasmic foci. Dysregulated L1 expression is associated with several human diseases - e.g., cancer, autoimmunity, neurodegeneration - and, generally, with aging. L1 therefore represents a promising biomarker opportunity. ORF2p is the 'business end' of L1, possessing the requisite endonuclease and reverse transcriptase activities, yet the mechanisms of L1 mobilization remain poorly understood. This is in-part because, even when L1s are naturally expressed, ORF2p is extremely lowly abundant.
The Research Specialist will be responsible for advancing our research team's L1 analyses - especially around the areas of L1 RNP composition and the in vitro activity of affinity enriched L1 RNPs. This will involve various biochemical and 'omic techniques. Working knowledge of mass spectrometry and hands-on experience with data collection and bioinformatic analyses is an advantage. The Research Specialist will run some of their own projects semi-independently, in consultation with their direct report and will also assist/support PhD-level researchers in their projects. The Research Specialist may also engage in some team management / administration tasks, but these will be minimal compared to hands-on research.

Qualifications
REQUIRED QUALIFICATIONS:
Bachelor's degree required
Minimum of 4 years relevant research experience in academic and/or industrial research settings
Extensive experience with protein-centric analyses including:
Immuno-precipitation
SDS-PAGE / general staining
Western blotting
General protein and nucleic acids biochemistry, theory and techniques
Mass spectrometry for proteomics, at least data analysis
Nucleic acids sequencing (short and long read), experimental design and data analysis
The ability to work (semi)independently to support PhD-level scientists
PREFERRED QUALIFICATIONS:
Knowledge and experience working with LINE-1 retrotransposons
Prior industry experience
Prior expreience with advanced protein research, including mass spectrometry, will be prioritized over sequencing
